Joseph first appears on the record when he was admitted an inhabitant of the Island of Aquidneck (at Portsmouth) in January 1639. However, he soon left Portsmouth for Newport, newly founded by his brother, John Clarke. He is said to have been one of the original members of the First Baptist Church of Newport in 1644. Joseph must've married at about this time, but nothing is known about his wife. He possibly married a Margaret as a second wife.
In November 1661, Tobias Saunders, Robert Burdick, and Joseph Clarke were arrested for taking possession of lands of Southertown. Southertown was the name given by Connecticut to the area called Misquamicutt (later Westerly) by the Rhode Islanders who were settling there. "Joseph Clark of Road Island was released by the Constable upon a very urgent occasion."
