Ambrose Fowler is supposed to be a son of William Fowler, one of the first magistrates of the New Haven Colony, and a first settler at Milford but this has not been proven. He appeared at Windsor, CT as early as 1640, where he was one of Rev. John Warham's church who came from Dorchester, MA, and located at Windsor in 1636. Ambrose was one of a committee in 1641, Hartford Colony, to settle the bounds of Uncoway and Poquonnuck. His name is often met with in the records of that town. He sold land at Windsor in 1671, soon after which he removed and settled with his children at Westfield, MA. In 1675 there is a notice of the burning of "Mr. Ambrose Fowler's house and barn" at Windsor. His will was made in 1692, and proved in 1706, in which he names seven children, all recorded at Windsor.
