According to the Ross Family Information page, both of the kits state that the ancestor of Daniel Ross (b. 1760) was "Hugh Ross III, Lord of Ross; b. 1735, Scotland; d. 1799, Scotland; m. Lady Janet; b. 1740, Scotland, d.1768, Scotland" however, no source is provided for that statement. It does not appear that any other participants are documented descendants of that line so at this point, the DNA cannot clearly be used to substantiate the claim. Daniel Ross was a child when he immigrated from Sutherlandshire, Scotland with his unnamed mother, who died aboard the ship.

In a letter written by Joshua Ross, a nephew of John Ross, he gives the biography of John Ross: "John Ross was born October 3, 1790. He died in Washington, D. C., August 1, 1866, while representing the Cherokee Nation. "He was a grandson of John McDonald, an Indian trader who was a member of Clan MacDonald, Inverness, Scotland. His grandmother was Anna Shorey, of the Cherokee Bird Clan. "The good influence of John McDonald saved many American lives during the Revolution, and after the war, from the hands of the Spanish and English and the Indian warriors in the valleys and mountains of Tennessee. "It is said that he saved the life of Daniel Ross, of Sutherlandshire, Scotland, who crossed the ocean when he was a boy. His mother died on the water and was buried at sea. The orphan boy landed at Baltimore, Md. When he was a little older he joined Mayberry's Trading Expedition and set out for the Tennessee country. Group G of the Rose Project (Rose of Kilravock) have been proven to be in Sub-Clade L21 and Group 1 of the Ross Project are a close genetic match to Group G (67-5 counting CDY as a 1 step mutations). The two Ross paternal lines shown as members of Cluster Two are, according to the genetic distance shown between them and proven L165+ MacLeod Haplotype, are almost certainly also L165+. The break in the Ross paternal line appears to have taken place in a generation near 1620 AD.

The Most Recent Common Ancestor of the two Ross participants was Owl Francis Peter Lymon Ross; b. 1817 Sequatchie Valley, TN, therefore the mutation at DYS 447 (from the Clan modal of "26" to the Ross "25") pre-dates 1817.

A 37 marker haplotype can mutate at any time but typically survives for approximately 150 to 350 years (mutation rates vary among different paternal lines). It appears that the Ross modal at DYS 570 may be "18" as opposed to the "17" of the Clan Haplotype, however, testing of the Ancestry kit would need to be done at this marker to verify this. It is possible that a random mutation took place in kit 145061 in the past four generations since the birth of Owl Francis Peter.

A 67 marker haplotype can mutate at any time but typically survives for approximately 105 to 210 years; of the markers that both have tested in the 38 - 67 group of markers, the two paternals lines have not shown any mutation. Both kits carry the "15" at DYS 446 which is the Clan MacLeod Modal at that marker (differing from L165 which is a "13").

It is recommended that Kit 145061 be tested for L165 to confirm the Cluster Two relationship.
